In vivo Gene Writing with engineered retrotransposons

Computational reconstruction, screening, and engineering of non-LTR retrotransposons uncovers elements, termed Gene Writers, that enable stable transgene integration in human cells using all-RNA delivery. After extensive characterization and optimization, Gene Writers is deployed to generate functional CAR-T cells in vivo in a humanized mouse model.
